Recovery grade of the properties of streptomycin resistant bacteria-passed phage by passing through normal bacteria

Abstract
The recovery grade, by passing through normal bacteria, of properties of the phage passed three times through streptomycin(SM) resistant bacteria was investigated. For this experiment, the Klebsiella phage(Kp phage), it¢¥s host cell, Klebsiella pneumoniae, type 2, (Kp) and SM resistant Kp(SMKp) were used. The following results were obtained.
The adsorptions of the phage passed three times through SMKp(SMKp-phage) to SMKp and Kp were similar to that of Kp phage.
If the SMKp-phage is passed three times through Kp, the plaque appearance in SMKp became more similar to that of Kp phage than SMKp-phage. However, in Kp, the SMKp-phage, Kp-passed SMKp-phage and Kp phage formed analogous plaques.
The halo plaque formation of the SMKp-phage became similar grade, against SMKp-phage, to that of Kp phage by passing three times through Kp.
The phage growth in SMKp and Kp was still similar, against Kp phage, to that of SMKp-phage even though the SMKp-phage is passed 16 times through Kp.
The antigenicity of the SMKp-phage passed 19 times or 21 times through Kp was much different from that of Kp phage against the SMKp-phage.
When the Kp passed, 21 times, SMKp-phage, SMKp-phage and Kp phage are neutralized by lower concentration of the anti-Kp phage serum, the initial lag in the reaction was appeared and it showed egually a theoretical two-hit curve appearance.
Both recoveries of the SMKp-phage from SMKp and Kp were similar as that of Kp phage.
The minimum inhibitory concentration of SM to the SMKp and Kp was 140 mg/ml and 0.02 mg/ml.
